V1aR, V2R, and the oxytocin receptors exist as homo- and hetero-dimers in transfected HEK293T cells. Each monomer has a similar propensity to form homo- and hetero-dimers, indicating that their relative expression levels may affect the ration of homo-/hetero-dimer. Immature forms of the receptors can be immunoprecipitated as homo- and hetero-dimers. In addition, ER fractions include homo- and hetero-dimers. These results suggest that the oligomerization processes take place early during biosynthesis. Agonists or antagonists binding do not affect the BRET signals. Therefore, the dimerization state of the receptors is not regulated by ligand binding once they have reached the cell surface. |
